目的:观察清上蠲痛合剂对偏头痛大鼠的三叉神经脊束核尾部C-fos蛋白表达的影响。方法:SD大鼠84只,雌雄各半,随机分为正常组(生理盐水剂量:与等效剂量等容积),模型组(生理盐水剂量:与等效剂量等容积),清上蠲痛合剂预防组[等效剂量,7.46 g/(kg·d)],清上蠲痛合剂高、中、低剂量治疗组[等效剂量,7.46 g/(kg·d)],舒马普坦组[等效剂量,4.5 mg/(kg·d),浓度是1 m L/mg],每组12只,采用硝酸甘油(GTN)皮下注射法建立偏头痛大鼠模型。预防组连续给药6天后造模用药。观察挠头、爬笼出现时间,采用免疫组化方法检测三叉神经脊束核尾部C-fos蛋白表达。结果:治疗组(高)30 min、60 min、90 min和120 min挠头次数显著低于模型对照组(P<0.05);治疗组(中)60 min和120 min挠头次数显著低于模型对照组(P<0.05);治疗组(低)60 min和120 min挠头次数显著低于模型对照组(P<0.05);治疗组(高)30 min、90 min和120 min爬笼次数显著低于模型对照组(P<0.05);治疗组(中)30 min、90 min和120 min爬笼次数显著低于模型对照组(P<0.05);阳性组、清上蠲痛合剂预防组、高、中剂量治疗组三叉神经脊束核C-fos阳性细胞数相对模型组有显著下降(P<0.05);清上蠲痛合剂预防组、治疗组高、治疗组中、阳性组组之间无统计学意义((P>0.05)。结论:清上蠲痛合剂能明显预防和改善大鼠偏头痛发作,可能与抑制三叉神经脊束核尾部C-fos蛋白表达有关。
Objective: To observe the effect of Qing Shang Tiao Tong Mixture on the expression of C-fos protein in the tail of the trigeminal nerve bundle of migraine rats. Methods: Eighty-four SD rats were randomly divided into normal group (saline dose and equivalent volume), model group (saline dose and equivalent volume) Preventive group [equivalent dose, 7.46 g / (kg · d)], Qing Shangchong Mixture high, medium and low dose treatment group [equivalent dose, 7.46 g / (kg · d)], sumatriptan group [Equivalent dose of 4.5 mg / (kg · d), concentration of 1 m L / mg] with 12 rats in each group. Migraine rat model was established by subcutaneous injection of nitroglycerin (GTN). Prevention group continuous administration of 6 days after modeling drug. The time of scratching the head and climbing the cage was observed. The expression of C-fos protein in the tail of the trigeminal nucleus was detected by immunohistochemistry. Results: The numbers of scratching at 30 min, 60 min, 90 min and 120 min in the treatment group were significantly lower than those in the model control group (P <0.05). The numbers of scratching at 60 min and 120 min in the treatment group were significantly lower than those in the model control group (P <0.05). The numbers of scratching scratches at 60 min and 120 min in the treatment group were significantly lower than those in the model control group (P <0.05). The numbers of climbing cage in the treatment group (30 min, 90 min and 120 min) (P <0.05). The numbers of climbing cage in the treatment group (middle) at 30 min, 90 min and 120 min were significantly lower than those in the model control group (P <0.05); The positive group, Qing Shang Tuo Tong Mixture prevention group, The number of C-fos-positive cells in the trigeminal nucleus of the trigeminal nerve decreased significantly in the dose-treated group compared with the model group (P <0.05). There was no significant difference between the two groups in the prevention and treatment group, the treatment group and the treatment group (P0.05) .Conclusion: Qing Shang Tiao Tong mixture can obviously prevent and improve migraine attack, which may be related to the inhibition of C-fos protein expression in the tail of the trigeminal nerve bundle.
