CD4 0 /CD4 0L相互作用在T细胞活化过程中扮演了十分重要的角色 ,参与了体内许多重要的生理和病理过程。本实验在建立了稳定表达CD4 0 Ig的CHO工程细胞株的基础上 ,对CHO工程细胞株的无血清大规模培养和融合蛋白的纯化进行了探索 ,并体外观察了纯化CD4 0 Ig对人CFU T的影响。结果表明 ,培养上清中融合蛋白产量约在 10 μg/ml以上 ,经ProteinG亲和凝胶纯化后蛋白纯度达 95 %以上。此外 ,该融合蛋白在体外可显著抑制人CFU T的形成 ,为进一步研究其体内功能奠定了良好基础 CD4 0 / CD4 0L interactions play an important role in the activation of T cells and are involved in many important physiological and pathological processes in the body.
