为了研究丝瓜提取物对衰老模型小鼠学习记忆能力和脑组织中氧自由基代谢及脑细胞膜离子转运的影响,试验将60只昆明种小鼠随机分为空白组、衰老模型组和丝瓜提取物高、中、低剂量组,通过皮下注射D-半乳糖制备衰老模型,8周后对各组小鼠行八臂迷宫行为学测试,测定小鼠脑组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性及丙二醛(MDA)的含量,脑组织Na+,K+-ATP酶和Ca2+,mg2+-ATP酶的活性及脑指数。结果表明:丝瓜提取物高、中剂量组衰老小鼠的短期记忆和长期记忆均有所改善;丝瓜提取物可提高衰老小鼠脑组织中抗氧化酶SOD、GSHPX活性,并明显降低MDA含量,提高脑组织中Na+,K+-ATP酶和Ca2+,mg2+-ATP酶活性。说明丝瓜提取物可改善衰老小鼠学习记忆能力,同时可通过增强机体清除氧自由基能力提高抗氧化酶活性及细胞膜蛋白活性而发挥抗衰老作用。
In order to study the effect of loofah extract on the learning and memory abilities of aging mice and the metabolism of oxygen free radicals in brain tissue and ion transport in brain cells, 60 Kunming mice were randomly divided into blank group, aging model group and loofah extract High, medium and low dose groups. The model of aging was established by subcutaneous injection of D-galactose. After 8 weeks, the mice in each group were subjected to eight-arm maze test to determine the superoxide dismutase (SOD) The activity of glutathione peroxidase (GSH-PX) and malondialdehyde (MDA), the activity of Na +, K + -ATPase and Ca2 +, mg2 + -ATPase in brain tissue and brain index were measured. The results showed that both the short-term memory and the long-term memory of senile mice in loofah extract group were improved. The loofah extract could increase the activities of antioxidant enzymes SOD and GSHPX in the brain tissue of aging mice and significantly reduce the content of MDA, Improve Na +, K + -ATPase and Ca2 +, mg2 + -ATPase activity in brain tissue. The results showed that loofah extract can improve the learning and memory ability of aged mice and at the same time exert the anti-aging effect by enhancing the ability of scavenging oxygen free radicals and enhancing the activities of antioxidant enzymes and cell membrane proteins.
