为了解EB病毒编码基因BHRF1的表达对鼻咽癌细胞抵抗放射线损伤能力的改变,构建携有BHRF1的高表达载体,并转染鼻咽癌细胞株CNE2细胞,观察转染前后的癌细胞在放射线照射后于裸鼠体内成瘤能力改变的情况。结果发现,在放射线照射前,BHRF1表达细胞在裸鼠体内形成肿瘤时间较晚,肿瘤重量较轻(P<0.01);而经放射线照射后,BHRF1表达细胞在裸鼠体内的成瘤能力强于对照组(P<0.01),经原位末端标记检查发现其凋亡指数小于对照组(P<0.01)。提示BHRF1的表达能通过阻止细胞的凋亡,增强鼻咽癌细胞抵抗放射线对其DNA的损伤作用。
To understand the effect of EBR-encoding gene BHRF1 on nasopharyngeal carcinoma cells against radiation-induced damage, a high expression vector carrying BHRF1 was constructed and transfected into CNE2 cells. After irradiation in nude mice tumor formation ability changes. The results showed that, before irradiation, BHRF1-expressing cells formed tumors in nude mice later, with lighter tumor weight (P <0.01). However, the BHRF1-expressing cells formed tumors in nude mice after irradiation (P <0.01). The apoptotic index was lower than that of the control group (P <0.01) by in situ terminal labeling. It is suggested that the expression of BHRF1 can enhance the DNA damage of nasopharyngeal carcinoma cells by preventing radiation by blocking the cell apoptosis.
