目的:探讨巴戟天提取物对D-半乳糖诱致小鼠学习记忆障碍的改善作用。方法:60只昆明种小鼠完全随机分为6组正常对照组、模型组、石杉碱甲组[0.4 mg/(kg·d)]、巴戟天提取物高剂量组[45g/(kg·d)],巴戟天提取物中剂量组[22.5 g/(kg·d)]和巴戟天提取物低剂量组[11.25 g/(kg·d)],每组10只。上述动物除正常对照组外,均皮下(sc)注射D-半乳糖8周,并于第5周起分别灌胃相应药物。采用水迷宫和自主活动仪测试小鼠学习和记忆能力;采用高效液相检测去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)二羟苯乙酸(DOPAC)的含量;测定血清糖化血红蛋白(Hb A1c)、果糖胺(FRA)、晚期糖基化终末产物(AGEs)和糖基化终末产物受体(RAGEs)以及醛糖还原酶活性水平(AR activity)。结果:巴戟天提取物明显改善模型小鼠学习和记忆能力;各给药组与模型对照组相比单胺类递质含量无明显变化;能明显降低药物处理后引起的AR活性增高,同时巴戟天提取物高、中、低剂量组均明显降低Hb A1c、FRA、AGEs和RAGEs。结论:巴戟天提取物具有改善D-半乳糖诱致小鼠学习记忆障碍的作用,其作用可能与其减少糖基化对脑神经细胞的损伤有关。
Objective: To investigate the effect of Morinda officinalis extract on D-galactose-induced learning and memory impairment in mice. Methods: Sixty Kunming mice were randomly divided into 6 groups: normal control group, model group, huperzine group [0.4 mg / (kg · d)], high dose of Morinda officinalis extract [45 g / (kg · D)], Morinda officinalis extract middle dose group [22.5 g / (kg · d)] and Morinda officinalis extract low dose group [11.25 g / (kg · d)]. In addition to the normal control group, the above animals were injected with D-galactose subcutaneously (sc) for 8 weeks, and the animals were respectively orally administered with the corresponding drugs from the 5th week onwards. The learning and memory abilities of mice were tested by water maze and autonomic activity meter. The levels of norepinephrine (NE), dopamine (DA), 5-hydroxytryptamine (5-HT) (HOPA), hydroxyproline (HOPA), diaphorase (DOPAC) were measured. Serum HbA1c, fructosamine, advanced glycation end products (AGEs) As well as aldose reductase activity level (AR activity). Results: Morinda officinalis extract significantly improved the ability of learning and memory in model mice. Compared with model control group, the content of monoamine neurotransmitters did not change obviously in each administration group, and the AR activity increased significantly after treatment Morinda officinalis extract high, medium and low dose groups were significantly lower Hb A1c, FRA, AGEs and RAGEs. Conclusion: Morinda officinalis extract has the effect of improving D-galactose-induced learning and memory impairment in mice, which may be related to the decrease of glycosylation on the damage of brain nerve cells.
