目的:研究蜀葵与药蜀葵特征DNA序列的差异,为鉴定维吾尔药材蜀葵及药蜀葵提供依据。方法:采用试剂盒法提取蜀葵及药蜀葵基因组DNA,PCR扩增ITS2片段,双向测序,应用Mega6.0软件分析序列,计算种内及种间遗传距离,构建NJ鉴别树。结果:蜀葵ITS2序列长度为233 bp,与药蜀葵231~232 bp存在差异;蜀葵种内最大K2P遗传距离为0.004,药蜀葵种内K2P遗传距离为0,蜀葵与药蜀葵种间平均K2P遗传距离为0.0732;NJ树结果表明蜀葵与药蜀葵均单独聚为一支。结论:ITS2序列可用于维吾尔药材蜀葵真伪鉴定的DNA条形码,为保障临床正确用药提供了新的方法。
Objective: To study the difference of characteristic DNA sequences of hollyhock and hollyhock, and to provide basis for identification of hollyhock and hollyhock. Methods: Genomic DNA of hollyhock and hollyhock were extracted by kit method. The ITS2 fragment was amplified by PCR and sequenced. The sequence was analyzed by Mega6.0 software, and the genetic distance between species and species was calculated to construct NJ discriminant tree. Results: The length of ITS2 sequence of Hollyhock was 233 bp, which was different from that of 231 ~ 232 bp. The maximum K2P genetic distance of Hollyhock was 0.004, the K2P genetic distance of Hollyhock was 0, the average K2P genetic distance between Hollyhye and Solanum glaucoides 0.0732; NJ tree results show that hollyhock and drug hollyhock are a single polygamy. Conclusion: ITS2 sequence can be used to identify DNA barcodes in hollyhock of Uyghur medicine, which provides a new method to ensure the correct clinical medication.
