目的:采用HPLC-ELSD方法测定不同产地欧亚旋覆花中1-O-乙酰旋覆花内酯的含量。方法:采用高效液相色谱法,蒸发光散射检测器。色谱柱:Diamonsil ODS C18(250 mm×4.6 mm,5μm);柱温:30℃;流动相:乙腈-水梯度洗脱,流速:0.8ml.min-1;ELSD条件:漂移管温度:100℃,载气流速:2.8 L.min-1。结果:1-O-乙酰旋覆花内酯线性范围为2.72~19.00μg,平均回收率98.91%,RSD=2.20%(n=6)。结论:方法可靠,简单可行,为控制中药材欧亚旋覆花的质量提供了依据。
OBJECTIVE: To determine the content of 1-O-acetolactone in Inula brazil flowers by HPLC-ELSD. Methods: High performance liquid chromatography and evaporative light scattering detector were used. Column: Diamonsil ODS C18 (250 mm × 4.6 mm, 5 μm); column temperature: 30 ° C .; mobile phase: acetonitrile-water gradient, flow rate: 0.8 ml.min-1; ELSD conditions: drift tube temperature: Carrier gas flow rate: 2.8 L.min-1. Results: The linear range of 1-O-acetolactone was 2.72 ~ 19.00μg with an average recovery of 98.91% and RSD = 2.20% (n = 6). Conclusion: The method is reliable, simple and feasible, and provides the basis for controlling the quality of Chinese medicine Euonymus japonicus.
