目的:通过油酸诱导肝癌HepG2细胞水通道蛋白9(AQP9)的表达观察后者对HepG2细胞株迁移能力的影响。方法:HepG2细胞经不同处理后(0、250、500μmol/L油酸处理或AgNO3+500μmol/L油酸处理),用Western blot法检测AQP9蛋白的表达,细胞划痕实验检测细胞迁移能力。结果:经油酸处理后,HepG2细胞AQP9蛋白表达增加,细胞迁移能力增强,且呈浓度依赖性(均P<0.05),油酸的上述作用被预先给予水通道蛋白抑制剂AgNO3所取消。结论:肝癌HepG2细胞的迁移能力与其AQP9表达有关,AQP9表达越高细胞的侵袭力越强。
OBJECTIVE: To observe the effect of oleic acid on the migration of HepG2 cells by observing the expression of aquaporin 9 (AQP9) in HepG2 cells. Methods: The HepG2 cells were treated with 0,250 and 500 μmol / L oleic acid or AgNO3 + 500 μmol / L oleic acid, respectively. Western blot was used to detect the expression of AQP9 protein. Cell scratch assay was used to detect cell migration. Results: After oleic acid treatment, the expression of AQP9 protein in HepG2 cells was increased, and the migration ability of HepG2 cells was enhanced in a concentration-dependent manner (all P <0.05). The above effect of oleic acid was abolished by pretreatment with aquaporin AgNO3. Conclusion: The migration ability of HepG2 cells is related to the expression of AQP9. The higher the expression of AQP9, the stronger the invasion of cells.
