目的使肺癌细胞表达共刺激分子,探讨其作为疫苗的可能性。方法小鼠骨髓细胞在体外经GM-CSF和TNF-α诱导,使之成为成熟树突状细胞(BmDC)。将其与小鼠Lewis肺癌细胞系AL9901融合,并以S-P免疫细胞化学染色法检测融合细胞和BmDC上有关分子的表达。结果通过GM-CSF和TNF-α诱导获得BmDC,以诱导5d时收获为佳。获得的BmDC能高表达B7-1和CD40分子,与肺癌细胞融合后,获得的融合细胞仍能高表达B7-1和CD40分子。结论通过将小鼠肺癌细胞与树突状细胞融合,使肺癌细胞获得了BmDC表达的B7-1和CD40分子,从而提高了其免疫原性,为制备肺癌疫苗奠定了基础。
Objective To enable lung cancer cells to express co-stimulatory molecules and discuss their potential as vaccines. Methods Mouse bone marrow cells were induced by GM-CSF and TNF-α in vitro to make mature dendritic cells (BmDC). It was fused with mouse Lewis lung cancer cell line AL9901 and the expression of the related molecules on the fusion cells and BmDC was detected by S-P immunocytochemistry staining. Results BmDC was induced by GM-CSF and TNF-α, and harvesting was better at 5 days. The obtained BmDC can highly express B7-1 and CD40 molecules. After fusion with lung cancer cells, the obtained fusion cells can still express high levels of B7-1 and CD40 molecules. Conclusion By fusion of mouse lung cancer cells with dendritic cells, lung cancer cells can obtain B7-1 and CD40 molecules expressed by BmDC, thus improving their immunogenicity and laying a foundation for the preparation of lung cancer vaccine.
