应用台盼蓝排染法 ,Wirght -Giemsa混合染色法 ,Hoechst3334 2荧光染色法及碘化丙啶(Propidiumiodide ,PI)与Hoechst 3334 2共染计数坏死细胞的PI阳性率测定法 ,研究 β -榄香烯吗素 (PIC -BE)及其与阿霉素 (ADM)联合应用对CEM/ADM细胞生长的影响。结果显示 :①PIC-BE能够显著抑制CEM/ADM细胞的生长 ,并诱导该细胞凋亡 ,其作用强度在一定范围内呈浓度和时间依赖性。②低毒剂量的PIC -BE与ADM联合应用可显著增强ADM对CEM/ADM细胞的生长抑制和凋亡诱导作用 ,其作用强度在一定范围内亦呈浓度和时间的依赖性。提示 :PIC -BE主要以诱导肿瘤细胞凋亡而表现其抗肿瘤作用 ,并且其对ADM的抗肿瘤作用具有协同效果
The PI-positive rate of necrotic cells was determined by trypan blue staining, Wirght-Giemsa mixed staining, Hoechst3334 2 fluorescence staining and Propidium iodide (PI) and Hoechst 3334 2 staining. Effect of Cardiotoxicamine (PIC -BE) and its combination with doxorubicin (ADM) on the growth of CEM / ADM cells. The results showed that: (1) PIC-BE could significantly inhibit the growth of CEM / ADM cells and induce the apoptosis of the cells. The intensity of action was in a concentration-dependent and time-dependent manner. The combination of low dose of PIC-BE with ADM could significantly enhance the growth inhibition and apoptosis induction of ADM on CEM / ADM cells in a concentration-dependent and time-dependent manner. Tip: PIC-BE mainly induces apoptosis of tumor cells to show its anti-tumor effect, and it has a synergistic effect on the anti-tumor effect of ADM
