目的:建立土荆芥中槲皮素和山柰素的含量测定方法。方法:用20%盐酸水解土荆芥中黄酮类成分得槲皮素和山柰素,并采用 HPLC 法测定槲皮素和山柰素的含量。色谱柱:Hypersil C_(18)(250 mm×4.6 mm,5 μm);流动相:乙腈-0.1%甲酸溶液(40:60);流速:1 mL·min~(-1);检测波长:360 nm。结果:槲皮素和山柰素的线性范围分别为0.0203~0.203μg(r=0.9999)及0.0944~0.944μg(r=0.9998);平均回收率(n=9)分别为99.3%及99.7%。结论:本法简单,重复性好,可用于土荆芥中槲皮素和山柰素含量的测定。
Objective: To establish a method for the determination of quercetin and kaempferol in earthworm Nepeta. Methods: Quercetin and kaempferol were obtained by hydrolyzing flavonoids from Nepeta serrata with 20% hydrochloric acid, and the contents of quercetin and kaempferol were determined by HPLC. Column: Hypersil C_(18)(250 mm×4.6 mm, 5 μm); mobile phase: acetonitrile-0.1% formic acid solution (40:60); flow rate: 1 mL·min~(-1); detection wavelength: 360 Nm. Results: The linear ranges of quercetin and kaempferol were 0.0203-0.203 μg (r=0.9999) and 0.0944-0.944 μg (r=0.9998), respectively; the average recoveries (n=9) were 99.3% and 99.7%, respectively. Conclusion: This method is simple and reproducible. It can be used to determine the content of quercetin and kaempferol in earthworm Nepeta.
