目的研究耐力运动和脂联素受体激动剂(AdipoRon)单独或联合干预对高脂饮食小鼠脂肪组织炎症的影响。方法 50只雄性C57BL/6小鼠随机分为普通饮食对照组(NC)、高脂饮食对照组(HC)、高脂饮食运动组(HE)、高脂饮食+AdipoRon组(HA)和高脂饮食+运动+AdipoRon组(HEA),每组10只。HE和HEA组进行6周中等强度耐力跑台训练;HA和HEA组按50 mg/kg剂量隔天皮下注射25%AdipoRon盐溶液。检测小鼠血清糖、脂代谢参数和炎症因子;腹腔注射葡萄糖耐量实验(IPGTT)检测小鼠糖耐量;RT-PCR法检测附睾脂肪基因表达。结果与NC组相比,HC组小鼠血清TG、TC、FFA、FBG和促炎因子TNF-α、IL-6浓度显著升高(P<0.05);TNF-α、IL-6、F4/80、CD11c、HIF-α、PDK1、GLUT1mRNA表达显著升高(P<0.05),IL-10 mRNA表达显著下降(P<0.05),糖耐量明显受损。HE和HA组小鼠除FBG、IL-10外,各项指标均显著高于NC组(P<0.05);与HC组相比,HE和HA组小鼠血清TG、TC、FFA、FBG、TNF-α、IL-6浓度和TNF-α、IL-6、F4/80、CD11c mRNA表达显著下降(P<0.05),IL-10表达显著升高(P<0.05),HIF-α、PDK1、GLUT1 mRNA表达下降无显著性差异;HEA组血清TG、FFA、TNF-α浓度和TNF-α、IL-6、F4/80、HIF-α、PDK1、GLUT1 mRNA表达较HE和HA组具有显著性差异(P<0.05),HE、HA和HEA组糖耐量得到显著改善。结论有氧运动和AdipoRon可能通过改善脂肪组织缺氧来降低高脂饮食小鼠脂肪组织巨噬细胞数量和M1态激活,降低脂肪组织炎症,同时改善葡萄糖耐受受损和脂代谢紊乱,并且联合干预效果优于单一干预效果。
Objective To investigate the effects of endurance exercise and AdipoRon alone or in combination on adipose tissue inflammation in high fat diet mice. Methods Fifty male C57BL / 6 mice were randomly divided into normal diet control group (NC), high fat diet control group (HC), high fat diet exercise group (HE), high fat diet + AdipoRon group (HA) Diet + exercise + AdipoRon group (HEA), 10 rats in each group. HE and HEA groups were subjected to 6 weeks of moderate-intensity endurance treadmill training; HA and HEA groups were subcutaneously injected with 25% AdipoRon salt solution at 50 mg / kg every other day. Serum glucose, lipid metabolism parameters and inflammatory factors were detected in mice. The glucose tolerance of mice was measured by intraperitoneal injection of glucose tolerance test (IPGTT). The expression of epididymal fat gene was detected by RT-PCR. Results Compared with NC group, the concentrations of TG, TC, FFA, FBG and TNF-α and IL-6 in HC group were significantly increased (P <0.05); the levels of TNF-α, IL-6 and F4 / 80, CD11c, HIF-α, PDK1 and GLUT1mRNA expression (P <0.05), IL-10mRNA expression decreased significantly (P <0.05), impaired glucose tolerance. The levels of TG, TC, FFA, FBG, IL-10 in HE and HA groups were significantly higher than those in NC group (P <0.05) The levels of TNF-α and IL-6 and the expressions of TNF-α, IL-6, F4 / 80 and CD11c mRNA were significantly decreased (P <0.05) , The expression of GLUT1 mRNA in HEA group was significantly lower than that in HE and HA groups, but the levels of TG, FFA, TNF-α and TNF-α, IL-6, F4 / 80, HIF- (P <0.05), and the glucose tolerance of HE, HA and HEA groups were significantly improved. Conclusion Aerobic exercise and AdipoRon may reduce adipose tissue macrophage and M 1 state activation, decrease adipose tissue inflammation, improve impaired glucose tolerance and dyslipidemia by improving hypoxia of adipose tissue, Intervention effect is better than a single intervention effect.
