目的考察毛冬青浸膏外用对糖尿病溃疡模型小鼠创面愈合率的影响,并初步探讨其可能的作用机理。方法采用链脲佐菌素结合高脂饲料诱导2型糖尿病模型,在此基础上复制皮肤溃疡模型,将模型小鼠随机分为胰岛素组(I),胰岛素+毛冬青低、中、高剂量(I+L,I+M,I+H)组和胰岛素+羟苯磺酸钙(I+CDC)组,分别给药;另取正常小鼠复制皮肤溃疡模型,涂抹等量生理盐水。以面积法测定各组的创面愈合率,并采用免疫组化法检测增殖细胞核抗原(PCNA)和第八因子相关抗原(FⅧ-RAg)的表达情况。结果给药结束后,与单纯溃疡组小鼠相比较,其他各组创面愈合率均显著降低,PCNA的阳性表达率均显著下调;除胰岛素+羟苯磺酸钙组外其余各组FⅧ-RAg的阳性表达率均显著下调;与单用胰岛素组比较,胰岛素联合毛冬青低、中、高剂量(I+L,I+M,I+H)组创面愈合率均显著增加,PCNA和FⅧ-RAg的阳性表达率显著升高。结论毛冬青外用对糖尿病溃疡具有较显著的修复作用,其作用可能与创面组织中PCNA和FⅧ-RAg的表达增加,从而促进表皮细胞增殖和新生血管的形成有关。
Objective To investigate the effect of Mao Dongqing Extract on healing rate of wounds in mice with diabetic ulcer and to explore its possible mechanism. Methods Streptozotocin and high-fat diet were used to induce type 2 diabetes mellitus. On the basis of this model, skin ulcers were duplicated. The model mice were randomly divided into insulin group (I), insulin + Mao Dongqing low, medium and high dose I + L, I + M, I + H) and insulin + calcium dobesilate (I + CDC) groups were given separately. Another normal mouse was used to duplicate the model of skin ulcer, and the same amount of saline was applied. The wound healing rate of each group was determined by area method. The expression of proliferating cell nuclear antigen (PCNA) and factor VIII-related antigen (FⅧ-RAg) were detected by immunohistochemistry. Results After the administration, wound healing rates of other groups were significantly lower than those of simple ulcer groups, and the positive rate of PCNA expression was significantly decreased. In addition to the insulin + calcium dobesilate group, the other groups FⅧ-RAg (I + L, I + M, I + H) group was significantly higher than that of single insulin group (P <0.05) RAg positive expression rate was significantly increased. Conclusion External application of Mao Dongqing can significantly repair diabetic ulcer, and its effect may be related to the increased expression of PCNA and FⅧ-RAg in wound tissues, which may promote the proliferation of epidermal cells and the formation of neovascularization.
