目的探讨自然杀伤T细胞(natural killer T cell,NKT)在原因不明习惯性流产(unexplained recurrent spontaneous abortion,URSA)发生机制中的作用。方法有URSA史患者50例,其中未孕者20例为URSA非孕组,难免流产者30例为UESA难免流产组,52例正常体检者中正常未生育者20例为正常未育组,正常妊娠孕妇20例为正常妊娠组,有正常生育史12例为正常生育组,采用流式细胞术检测各组外周血中CD3+CD16+CD56+NKT细胞的表达水平。结果外周血CD3+CD16+CD56+NKT细胞百分率正常未生育组[(4.62±1.38)%]与正常生育组[(4.90±3.00)%]比较差异无统计学意义(P>0.05);URSA非孕组外周血CD3+CD16+CD56+NKT细胞百分率[(2.42±1.52)%]低于正常生育组(P<0.05),URSA难免流产组[(4.18±2.77)%]低于正常妊娠组[(14.40±2.27)%](P<0.05);正常妊娠组外周血中NKT细胞百分率是正常生育组的3倍,而URSA难免流产组是URSA非孕组的2倍。结论 URSA患者外周血NKT细胞表达水平降低,NKT细胞可能参与了URSA的发生。
Objective To investigate the role of natural killer T cell (NKT) in the pathogenesis of unexplained recurrent spontaneous abortion (URSA). Methods There were 50 patients with URSA history, of which 20 were non-pregnant URSA non-pregnant group, 30 were inevitable abortion of UESA inevitable abortion group, 52 normal healthy subjects without normal fertility in 20 cases of normal non-fertility group, normal Twenty pregnant women were normal pregnancy group, 12 cases with normal birth history were normal fertility group. The expression of CD3 + CD16 + CD56 + NKT cells in peripheral blood of each group was detected by flow cytometry. Results There was no significant difference in the percentage of CD3 + CD16 + CD56 + NKT cells in normal non-fertile group [(4.62 ± 1.38)%] vs normal fertile group (4.90 ± 3.00%) (P> 0.05) The percentage of CD3 + CD16 + CD56 + NKT cells in the pregnant group was significantly lower than that in the normal pregnancy group [(2.42 ± 1.52)%] [(4.18 ± 2.77)%] in the URSA inevitable abortion group [ (14.40 ± 2.27)%] (P <0.05). The percentage of NKT cells in peripheral blood of normal pregnant women was 3 times higher than that of normal pregnant women, but it was twice as that of URSA non-pregnant women. Conclusion The expression of NKT cells in peripheral blood of URSA patients is decreased, and NKT cells may be involved in the pathogenesis of URSA.
