应用胶原酶消化法获取人脐带静脉内皮细胞,在体外传代培养.以 HFRS 病毒感染培养的人脐带静脉内皮细胞做为模型,采用刚果红染色法在感染后的12小时、3天、5天分别检测与 PHA 活化淋巴细胞及 TNF—α刺激嗜中性粒细胞的粘附作用.发现在第3天、第5天感染内皮细胞与上述两种细胞的粘附作用明显高于未感染的内皮细胞,提示 HFRS 病毒的感染可能促进了人脐带静脉内皮细胞粘附分子的表达.
Human umbilical vein endothelial cells were obtained by collagenase digestion and subcultured in vitro.With human umbilical vein endothelial cells (HFU) infected by HFRS virus as a model, the cells were harvested at 12, 3 and 5 days after infection with Congo red staining The adhesion of endothelial cells to PHA-activated lymphocytes and TNF-α-stimulated neutrophils was measured and it was found that the adhesion of endothelial cells to both cells on day 3 and 5 was significantly higher than that of non-infected endothelial cells , Suggesting that HFRS infection may promote the expression of human umbilical vein endothelial cell adhesion molecules.
