目的测定芒果苷在正辛醇-水系统中的油水分配系数,考察系统中pH对油水分配系数的影响。方法采用HPLC法测定芒果苷的含量,并采用摇瓶法测定芒果苷在不同pH的正辛醇-水中的油水分配系数。结果芒果苷在pH为1.32、2.52、3.32、4.50、6.86、7.40、8.01时,油水分配系数平均值分别为2.234、2.313、2.125、1.907、0.246、0.008、0.005。结论 pH可显著影响芒果苷的油水分配系数,在胃肠环境中油水分配系数很小,可能是造成芒果苷在胃肠道全段通透性较差的原因之一。
Objective To determine the oil-water partition coefficient of mangiferin in n-octanol-water system and investigate the effect of pH on the oil-water partition coefficient. Methods The content of mangiferin was determined by HPLC. The oil-water partition coefficient of mangiferin in n-octanol-water with different pH was determined by shake flask method. Results The average oil-water partition coefficients of mangiferin at pH 1.32,2.52,3.32,4.50,6.86,7.40,8.01 were 2.234,2.313,2.125,1.907,0.246,0.008,0.005, respectively. Conclusion pH can significantly affect the oil-water partition coefficient of mangiferin, and its oil-water partition coefficient is very small in the gastrointestinal environment, which may be one of the reasons for the poor permeability of mangiferin in the whole gastrointestinal tract.
