背景:地氟烷可减少大鼠不完全脑缺血模型神经功能的缺失,然而,形态学上地氟烷脑保护作用的证据尚不充足。目的:研究地氟烷对全脑缺血再灌注损伤大鼠超微结构的影响,探讨地氟烷的脑保护作用。设计:随机对照的实验研究。单位:首都医科大学附属北京天坛医院。材料:在首都医科大学附属北京神经外科研究所对9只Wistar大鼠进行研究。随机分为3组,缺血组(n=3),地氟烷组(n=3)和假手术对照组(n=3)。干预:制备大鼠全脑缺血再灌注模型。地氟烷组再灌注开始后立即吸入地氟烷1h。缺血组和地氟烷组于缺血再灌注1h、假手术组于手术后1h取脑,利用电镜技术观察皮质超微结构的变化。主要观察指标:皮质超微结构变化。结果:与缺血组比较,地氟烷组神经元固缩小,神经元的细胞器和微管结构基本正常,但胶质细胞和微循环的破坏并无改善。结论:地氟烷对神经元、细胞骨架可能具有保护作用,但对胶质细胞和微循环可能无保护作用。
BACKGROUND: Desflurane reduces the loss of neurological function in incomplete cerebral ischemia in rats. However, evidence of desflurane brain protection is not yet sufficient. Objective: To study the effects of desflurane on ultrastructure in rats with global cerebral ischemia-reperfusion injury and to explore the neuroprotective effect of desflurane. Design: Randomized controlled experimental study. SETTING: Beijing Tiantan Hospital Affiliated to Capital Medical University. MATERIALS: Nine Wistar rats were studied at the Beijing Institute of Neurosurgery, Capital Medical University. The rats were randomly divided into 3 groups: ischemia group (n = 3), desflurane group (n = 3) and sham operation control group (n = 3). Intervention: Preparation of rat global cerebral ischemia-reperfusion model. Desflurane inhaled desflurane immediately after reperfusion began 1h. The ischemic group and the desflurane group in ischemia reperfusion 1h, sham operation group at 1h after surgery to take the brain, using electron microscopy to observe changes in cortical ultrastructure. MAIN OUTCOME MEASURES: Changes in cortical ultrastructure. Results: Compared with the ischemic group, the neurons in desflurane group showed a little shrinkage, the organelles and microtubules of neurons were basically normal, but the destruction of glial cells and microcirculation did not improve. CONCLUSION: Desflurane may have protective effects on neurons and cytoskeleton, but may not protect glial cells and microcirculation.
