[目的]探讨白介素-17(IL-17)mRNA及干扰素-γ(IFN-γ)mRNA在内毒素诱导性葡萄膜炎大鼠模型中的表达及其意义.[方法]将40只SD大鼠分为空白对照组(n=10)与模型组(n=30),模型组均采用内毒素诱导葡萄膜炎大鼠模型,并在注药6h、12h、18h、24h、48h、60h分别处死5只大鼠,对照组注射磷酸缓冲盐溶液(PBS)24h后处死10只,均取双眼葡萄膜视网膜组织,采用反转录聚合酶链式反应法(RT-PCR)检测两组眼部组织IL-17mRNA及IFN-γmRNA水平,显微镜下观察眼部组织炎症情况并按相关标准予以评分.[结果]①模型组造模不同时间眼部炎症评分均高于空白对照组(P<0.05),模型组造模6h、12h、18h后炎症评分依次升高(P<0.05),18h到达峰值,然后24h、48h、60h依次降低(P<0.05);②模型组造模不同时间IL-17 mRNA、IFN-γ mRNA水平均高于空白对照组(P<0.05),模型组造模6h、12h、18h后IL-17mRNA、IFN-γ mRNA水平依次升高(P<0.05),18h到达峰值,然后24h、48h、60h依次降低(P<0.05).[结论]葡萄膜炎大鼠眼部组织中IL-17 mRNA、IFN-γ mRNA表达水平上调,与病变程度呈一致,IL-17、IFN-γ均可能参与葡萄膜炎的发病过程.“,”[Objective]To analyze the expression of interleukin-17(IL-17)and interferon gamma(IFN-γ)messenger RNA(mRNA)in eyes of rats with endotoxin induced uveitis.[Methods] Forty SD rats were randomly divided into the blank control group(n =10)and the model group(n =30).Rats in the model group were treated with endotoxin to in-duce uveitis,and 5 rats were sacrificed at 6 h,12 h,18 h,24 h,48 h,60 h,respectively;the control group was injected with phosphate buffered saline(PBS),and 10 rats were sacrificed after 24h.Uveal retina of all of them were taken from the both eyes.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect the levels of IL-17 mRNA and IFN-γ mRNA of ocular tissues in the two groups.The inflammation of the eye tissue was observed under microscope and scored according to relevant criteria.[Results] ① The ocular inflammation score in the model group at different time was higher than that in blank control group(P<0.05);in the model group,the inflammatory scores were increased suc-cessively after 6 h,12 h and 18 h(P<0.05),and reached the peak at 18 h,then decreased successively at 24 h,48 h, 60 h(P<0.05).② The IL-17 mRNA and IFN-γ mRNA levels in the model group at different times were higher than those in the blank control group(P<0.05);levels of IL-17mRNA and IFN-γ mRNA in the model group at 6 h,12 h, 18 h increased successively(P<0.05),and reached the peak at 18 h,then successively decreased at 24 h,48 h,60 h (P<0.05).[Conclusion]The expression levels of IL-17 mRNA and IFN-γ mRNA in the ocular tissues of uveitis rats are up regulated,which are consistent with the extent of lesions;IL-17,IFN-γ may be involved in the pathogenesis of uveitis.
