目的 :探讨孕酮 (Progesterone ,Prog)及地塞米松 (Dexamethasone,Dex )对人外周血T淋巴细胞体外活化CD6 9表达的作用。方法 :以女性健康志愿者 (10名 )为研究对象 ,以蛋白激酶C的刺激剂佛波醇酯 (Phorbolester,PDB)为T淋巴细胞的活化剂 ,采用双荧光染色流式细胞技术 ,检测CD3+的T淋巴细胞表达早期活化表面分子CD6 9的百分率 ,以观察Prog、Dex及Prog加上Dex对外周血T淋巴细胞体外活化的作用。结果 :在体外培养条件下 ,无论单独使用Prog还是Dex均增强低剂量PDB刺激CD6 9的表达。然而 ,与单独使用Prog或Dex的作用相比 ,Prog加上Dex明显减弱低剂量PDB的活化CD6 9的表达。结论 :同时使用Prog与Dex可明显抑制T淋巴细胞的体外活化 ,提示在临床上联合使用Prog和Dex对自身免疫性疾病病人可能产生抑制免疫炎症作用
Objective: To investigate the effect of Progesterone (Prog) and Dexamethasone (Dex) on the activation of CD6 9 in human peripheral blood T lymphocytes in vitro. Methods: A total of 10 healthy female volunteers were enrolled in this study. Phospholipase stimulator Phorbolide (PDB) was used as an activator of T lymphocytes. Double fluorescent staining was used to detect CD3 + Of T lymphocytes express early activated surface molecule CD6 9 in order to observe the effect of Prog, Dex and Prog plus Dex on the activation of peripheral blood T lymphocytes in vitro. RESULTS: Under in vitro culture conditions, either Prog or Dex alone enhanced low-dose PDB to stimulate CD6 9 expression. However, Prog plus Dex significantly attenuated the expression of activated CD69 of low dose PDB compared to the effect of Prog or Dex alone. Conclusions: Prog and Dex can inhibit the activation of T lymphocytes in vitro, suggesting that the combination of Prog and Dex can inhibit the immune inflammation in patients with autoimmune diseases
