目的:观察中药掌叶半夏提取物的有效部位(PE)对CaSki和HeLa细胞人乳头瘤病毒(HPV)E6基因和P53基因表达的影响,进一步阐明PE的抑癌机制。方法:0.15mg/mL的PE作用于CaSki和HeLa细胞24h后,采用RT-PCR方法检测两种细胞HPVE6和P53 mRNA表达的变化,采用Western Blot检测CaSki和HeLa细胞HPVE6、P53蛋白表达的变化。结果:0.15mg/mL的PE作用24h后CaSki和HeLa细胞HPVE6 mRNA和蛋白的表达均明显下降,而P53mRNA和蛋白的表达均明显上升(P<0.05,P<0.01)。结论:PE可以明显抑制CaSki和HeLa宫颈癌细胞株HPVE6基因的表达、促进其P53基因的表达,这可能是PE抗宫颈癌作用的关键环节。
OBJECTIVE: To observe the effects of the extract of Pinellia ternata extract (PE) on the E6 gene and P53 gene expression of human papillomavirus (HPV) in CaSki and HeLa cells and further elucidate the mechanism of PE. Methods: After treated with 0.15mg / mL PE for 24 hours, the expressions of HPVE6 and P53 mRNA in CaSki and HeLa cells were detected by RT-PCR. The expression of HPVE6 and P53 in CaSki and HeLa cells was detected by Western Blot. Results: The expression of HPVE6 mRNA and protein in CaSki and HeLa cells was significantly decreased after treated with 0.15mg / mL PE for 24 h, while the expression of P53 mRNA and protein were significantly increased (P <0.05, P <0.01). Conclusion: PE can significantly inhibit the expression of HPVE6 gene in CaSki and HeLa cervical cancer cell lines and promote the expression of P53 gene, which may be the key point of PE anti-cervical cancer.
