目的:研究温胆汤对精神分裂症模型大鼠行为学及其海马组织神经调节蛋白1(NRG1)、ErbB4 mRNA表达的影响。方法:将60只大鼠随机分为6组,即正常组、模型组、氯氮平20 mg/kg组、温胆汤40g/kg组、温胆汤20g/kg组、温胆汤10g/kg组。正常组和模型组分别ig生理盐水20 ml/kg,氯氮平20 mg/kg组ig氯氮平原药20 mg/kg,温胆汤40、20、10 g/kg组分别ig温胆汤生药40,20,10 g/kg,1次/d,共21 d。在末次给药2 h后,除正常组外,其余各组一次性ip MK-801 0.6 mg/kg,以诱发精神分裂症模型,造模后,观察并记录各组大鼠的行为学改变,观察3 d后,处死并取海马组织检测,观察大鼠的行为学改变,并做刻板行为和共济失调评分;RT-PCR检测大鼠海马组织NRG1、ErbB4 mRNA表达。结果:与正常组比较,模型组大鼠刻板行为和共济失调评分明显升高;其海马组织NRG1、ErbB4 mRNA的表达也明显升高。与模型组比较,温胆汤40、20、10 g/kg组均能明显降低大鼠刻板行为和共济失调评分;也能明显降低海马组织NRG1、ErbB4 mRNA的表达。结论:温胆汤通过降低精神分裂症易感基因NRG1及其受体ErbB4蛋白的表达,改善了大鼠的行为学改变,从而防治精神分裂症。
Objective: To investigate the effect of Wendan Decoction on behavior and the expression of NRG1 and ErbB4 mRNA in hippocampus of schizophrenic model rats. Methods: 60 rats were randomly divided into 6 groups: normal group, model group, clozapine 20 mg / kg group, Wendan Decoction 40g / kg group, Wen Dan Tang 20g / kg group. The normal group and the model group were given ig saline 20 ml / kg, clozapine 20 mg / kg group ig clozapine 20 mg / kg, Wen Dan Tang 40,20,10 g / 40,20,10 g / kg, 1 time / d, a total of 21 d. After the last administration for 2 h, except for the normal group, all the other groups were given ip MK-801 0.6 mg / kg once to induce schizophrenia model. After modeling, the behavioral changes of each group were observed and recorded. After 3 days of observation, the rats were sacrificed and the hippocampus tissues were taken for detection. The behavioral changes of the rats were observed, and stereotyped behavior and ataxia score were performed. The expressions of NRG1 and ErbB4 mRNA in the hippocampus were detected by RT-PCR. Results: Compared with the normal group, the stereotypic scores of ataxia and ataxia in the model group were significantly increased. The expressions of NRG1 and ErbB4 mRNA in the hippocampus were also significantly increased. Compared with the model group, Wen Dan Tang 40,20,10 g / kg group can significantly reduce the stereotyped behavior and ataxia score; also significantly reduce NRG1, ErbB4 mRNA expression in the hippocampus. Conclusion: Wendan Decoction can prevent schizophrenia by decreasing the expression of NRG1 and its receptor ErbB4 in schizophrenia and improving the behavioral changes in rats.
