目的:研究人类疱疹病毒6型(HHV-6)感染与口腔鳞癌的相关性。方法:用PCR和间接免疫荧光方法分别检测口腔鳞癌患者和健康志愿者唾液HHV-6DNA和血清抗HHV-6IgG抗体。免疫组化法检测口腔鳞癌组织及癌旁组织标本中HHV-6糖蛋白B(glycoprotein B,gB)抗原。结果:118份口腔鳞癌唾液标本中PCR阳性率为65.3%,高于正常人群的40%(P<0.05)。HHV-6感染与患者病历资料无显著相关性。18份口腔鳞癌患者和20份健康人血清中HHV-6IgG抗体的几何平均滴度分别为1∶102.17和1∶101.50(P<0.05)。免疫组化法检测,11例口腔鳞癌标本中6例出现的阳性结果,胞质内出现弥散性的黄色颗粒,在癌巢内特异性浓集,而间质为阴性。8例相应癌旁组织中3例出现阳性染色结果,但阳性强度较癌组织弱且弥散。1例gB组化阳性肿瘤组织细胞经分离后接种裸鼠,移植瘤组织也检测到HHV-6抗原。结论:本研究说明HHV-6与口腔鳞癌的发生存在相关性,但不与患者临床资料分布相关。本研究亦提示荷瘤裸鼠作为HHV-6动物模型的可能性。
Objective: To study the correlation between human herpesvirus type 6 (HHV-6) infection and oral squamous cell carcinoma. Methods: Salivary HHV-6 DNA and anti-HHV-6 IgG were detected by PCR and indirect immunofluorescence in oral squamous cell carcinoma patients and healthy volunteers respectively. Immunohistochemistry was used to detect the antigen of HHV-6 glycoprotein B (gB) in oral squamous cell carcinoma tissues and adjacent tissues. Results: The positive rate of PCR in saliva samples from 118 oral squamous cell carcinomas was 65.3%, higher than that of normal population (P <0.05). There was no significant correlation between HHV-6 infection and patient’s medical records. The geometric mean titers of HHV-6 IgG antibodies in sera of 18 oral squamous cell carcinomas and 20 healthy volunteers were 1:102.17 and 1:101.50, respectively (P <0.05). Immunohistochemical detection, 11 cases of oral squamous cell carcinoma specimens of positive results occurred in 6 cases, the cytoplasm appears diffuse yellow granules, specifically concentrated in the cancer nest, and interstitial negative. In 8 cases of corresponding paracancerous tissues, positive staining results were observed in 3 cases, but the positive staining intensity was weaker than that in the cancerous tissues. One case of gB-positive tumor cells were isolated and inoculated nude mice, HHV-6 antigen was also detected in the xenografts. Conclusion: This study shows that HHV-6 is associated with the occurrence of oral squamous cell carcinoma, but not with the distribution of clinical data. This study also suggests the possibility of tumor-bearing nude mice as an animal model of HHV-6.
