利用人卵巢癌细胞株,在确定C-erbB-2扩增和过表达及C-erbB2正常拷贝和低表达的基础上对C-erbB-2扩增及表达与TNF和LAK杀伤活性的关系进行研究,并以非特异性杀伤物H2O2为对照。结果表明:C-erbB-2扩增及过表达可明显抑制TNF和LAK的杀伤活性(P<0.05),但对H2O2的杀伤活性无显著影响。提示临床上部分卵巢癌患者用TNF和LAK治疗的疗效低或无反应性与C-erbB-2的扩增及过表达有关。
Using human ovarian cancer cell lines, the relationship between C-erbB-2 amplification and TNF and LAK killing activity was determined on the basis of confirming C-erbB-2 amplification and overexpression and normal copy and low expression of C-erbB2 Study, and non-specific killing H2O2 as a control. The results showed that C-erbB-2 expansion and overexpression could significantly inhibit the cytotoxic activity of TNF and LAK (P <0.05), but had no significant effect on the cytotoxic activity of H2O2. Prompt clinical ovarian cancer patients with TNF and LAK treatment of low or no response with the C-erbB-2 amplification and overexpression.
