目的:探讨红景天苷对MPP+诱导的PC12细胞凋亡是否具有保护作用。方法:采用MPP+诱导的具有多巴胺能神经元特性的PC12细胞凋亡作为PD的体外模型。实验分对照组、500μmoL/L MPP+诱导组、红景天苷(10、50、100μmol/L)3个浓度预处理组。用MTT法测定细胞活性,流式细胞术检测细胞凋亡,TUNEL法检测细胞凋亡断裂的DNA片段。结果:10、50、100μmoL/L红景天苷对MPP+诱导的PC12细胞具有一定的保护作用。与MPP+组(50.2±1.8)%相比,10、50、100μmoL/L红景天苷预处理细胞活力分别上升为(65.1±1.0)%、(69.5±2.3)%、(80.9±2.0)%(P<0.05)。流式细胞术检测提示正常组、MPP+组、红景天苷预处理组(10、50、100μmoL/L)凋亡百分率分别为0.9%、34.5%、26.9%、20.1%、14.2%。经红景天苷预处理后,与MPP+组相比较细胞断裂的DNA片段明显减少。结论:红景天苷对MPP+诱导的细胞凋亡具有浓度依赖性的保护作用。
Objective: To investigate whether salidroside has a protective effect on apoptosis of PC12 cells induced by MPP+. METHODS: Apoptosis of PC12 cells with dopaminergic neuron characteristics induced by MPP+ was used as an in vitro model of PD. The experiment was divided into control group, 500μmoL/L MPP+ induction group, and salidroside (10, 50, 100μmol/L) pretreatment group. The cell viability was measured by MTT assay, cell apoptosis was detected by flow cytometry, and DNA fragmentation of apoptosis was detected by TUNEL assay. Results: 10, 50, and 100 μmol L/L salidroside had a protective effect on MPP+-induced PC12 cells. Compared with (50.2±1.8)% in MPP+ group, the viability of pretreated cells with 10, 50, and 100 μmol/L salidroside increased to (65.1±1.0)%, (69.5±2.3)%, and (80.9±2.0)%, respectively. (P<0.05). Flow cytometry indicated that the percentage of apoptotic cells in the normal group, MPP+ group, and salidroside pretreatment group (10, 50, 100 μmoL/L) were 0.9%, 34.5%, 26.9%, 20.1%, and 14.2%, respectively. After pretreatment with salidroside, the fragmented DNA fragments were significantly reduced compared to the MPP+ group. CONCLUSION: Salidroside has a concentration-dependent protective effect on MPP+-induced apoptosis.
