目的探讨外周血单个核细胞(PBMCs)干扰素诱导的蛋白10(IP-10)mRNA 表达与系统性红斑狼疮(SLE)活动的关系。方法用逆转录-聚合酶链反应(RT-PCR)检测46例 SLE 患者、20例类风湿关节炎(RA)患者、11例非 SLE 肾损伤患者及20名正常健康人 PBMCs IP-10 mRNA 表达。结果 SLE 活动组 PBMCs IP-10 mRNA 表达水平与非活动组间的差异有统计学意义(P<0.01),活动性狼疮肾炎(LN)组与无肾损伤组之间 PBMCs IP-10 mRNA 表达水平的差异亦有统计学意义(P<0.05),而 RA 组和非 SLE 肾病组患者与正常对照组水平相近似(P>0.05)。血清 IP-10水平与PBMCs IP-10 mRNA 表达高度正相关(r=0.8971,P<0.01)。PBMCs IP-10 mRNA 表达水平与狼疮病活动指数(SLEDAI)评分呈正相关(r=0.683 7,P<0.01),与肾 SLEDAI 评分亦高度正相关(r=0.726 0,P<0.01)。结论 SLE 活动时期 PBMCs IP-10 mRNA 表达增加,与 SLE 疾病活动状况相关,尤其与 LN 的活动状况密切相关。
Objective To investigate the relationship between interferon-induced protein 10 (IP-10) mRNA expression in peripheral blood mononuclear cells (PBMCs) and systemic lupus erythematosus (SLE) activity. Methods The expressions of IP-10 mRNA in 46 SLE patients, 20 rheumatoid arthritis (RA) patients, 11 non-SLE renal injury patients and 20 normal healthy people PBMCs were detected by reverse transcription-polymerase chain reaction (RT-PCR) . Results The expression of IP-10 mRNA in PBMCs in SLE group was significantly lower than that in non-active group (P <0.01). The expression of IP-10 mRNA in PBMCs in active lupus nephritis (LN) group and non-renal injury group (P <0.05), while the levels of RA and non-SLE nephropathy group were similar to those of the normal control group (P> 0.05). Serum IP-10 level was positively correlated with the expression of IP-10 mRNA in PBMCs (r = 0.8971, P <0.01). The expression of IP-10 mRNA in PBMCs was positively correlated with SLEDAI score (r = 0.683 7, P <0.01), and positively correlated with SLEDAI score (r = 0.726 0, P <0.01). Conclusion The expression of IP-10 mRNA in PBMCs during SLE is increased, which is related to the activity of SLE, especially closely related to the activity of LN.
