[目的]筛选降解小麦秸秆纤维素的真菌并分析C/N比对纤维素酶活力的影响。[方法]应用刚果红鉴定培养基和滤纸条降解度分析法筛选菌株,通过比对真菌rDNA的ITS区域序列鉴定菌株类型,通过调节培养基葡萄糖和(NH4)2SO4的比例研究了纤维素降解酶活性。[结果]从土壤中筛选出有2株分解纤维素能力较强的真菌,分别命名为NY01和NY02;真菌ITS保守序列比对结果显示,NY01与木霉的相似性最高达99%,NY02与毛霉的相似性高达99%;培养基中C/N比值在8∶1时2个菌株的CMC和FPA酶活性均达到最高。[结论]该研究为进一步探索秸秆纤维素降解奠定了基础。
[Objective] The study aimed to screen the fungi that degraded wheat straw cellulose and analyze the effect of C / N ratio on cellulase activity. [Method] The strain was screened by Congo red identification medium and filter paper degradation degree analysis. The strain type was identified by ITS sequence of fungal rDNA. The degradation of cellulose was studied by adjusting the ratio of glucose and (NH4) 2SO4 in medium. Enzyme activity. [Result] Two strains of fungi with strong ability of decomposing cellulose were screened out from the soil and named as NY01 and NY02, respectively. The comparison of ITS conserved sequence showed that the similarity between NY01 and Trichoderma was up to 99%, NY02 and The similarity of Mucor was up to 99%. The CMC and FPA activities of the two strains reached the highest at 8: 1 C / N ratio in the medium. [Conclusion] This study laid the foundation for further exploration of straw cellulose degradation.
