目的:以人的类成骨细胞株 T E85 和 U2 为细胞模型,观察雌激素对细胞功能的影响。方法:3 H胸腺嘧啶参入法测细胞增殖; E L I S A 法测细胞因子 I L6 含量;精氨酸转化法测i N O S 活性。结果: I L1(20 U·m L- 1 ) 和 T N Fα(50 U·m L- 1 ) 刺激细胞分泌 I L6 ,并抑制细胞增殖。在 T E85 细胞,17β雌二醇( E2 ) 可抑制 I L6 分泌并能对抗 I L1和 T N Fα对细胞增殖的影响。 N O S 抑制剂 L N M M A 使细胞3 H胸腺嘧啶的参入降低, E2 (1 n mol· L- 1) 可对抗 L N M M A 的抑制作用。结论: E2 通过减少 I L6 分泌和影响i N O S 活性而调节细胞功能。
OBJECTIVE: To investigate the effect of estrogen on cell function by using human osteoblast-like cell lines T E85 and U2 as cell models. Methods: 3 H thymidine incorporation assay cell proliferation; E L I S A method for the determination of cytokines I L 6 content; arginine conversion assay i N O S activity. RESULTS: I L1 (20 U · m L -1) and T N Fα (50 U · m L -1) stimulated cells to secrete I L6 and inhibited cell proliferation. In T E85 cells, 17β-estradiol (E2) inhibits the secretion of IL-6 and can counteract the effects of IL-1 and TNFα on cell proliferation. N O S inhibitor LN M M A reduced 3H-thymidine incorporation and E2 (1 n mol · L -1) inhibited LN M M A inhibition. Conclusion: E2 regulates cell function by decreasing I L-6 secretion and affecting i N O S activity.
