目的:研究雷公藤内酯醇对体外培养的人Th17细胞分化的调节作用。方法:在体外将不同浓度雷公藤内酯醇作用于人外周血单一核细胞、纯化的CD4+T细胞和CD4+CD45RA-记忆性T细胞,采用MTT法检测淋巴细胞增殖,流式细胞术分析单个细胞内表达IL-17和IFN-γ的频数,ELISA法测定细胞分泌IL-17和IFN-γ的水平。结果:雷公藤内酯醇以剂量依赖的方式抑制淋巴细胞增殖、细胞内IL-17的表达频数和细胞产生IL-17的水平。此外,雷公藤内酯醇对细胞内IFN-γ的表达和产生也有抑制作用。结论:雷公藤内酯醇可抑制人Th17细胞的分化,这可能是雷公藤内酯醇免疫抑制和抗炎作用的机制之一。
AIM: To investigate the regulation of triptolide on the differentiation of human Th17 cells cultured in vitro. Methods: Different concentrations of triptolide were administered to human peripheral blood mononuclear cells, purified CD4 + T cells and CD4 + CD45RA-memory T cells in vitro. The proliferation of lymphocytes was detected by MTT assay. Flow cytometry analysis of single The frequency of IL-17 and IFN-γ expression in cells was measured by ELISA. The levels of IL-17 and IFN-γ secreted by cells were measured by ELISA. Results: Triptolide inhibited lymphocyte proliferation, the frequency of intracellular IL-17 expression and the level of IL-17 produced by cells in a dose-dependent manner. In addition, triptolide also has an inhibitory effect on the expression and production of intracellular IFN-γ. Conclusion: Triptolide can inhibit the differentiation of human Th17 cells, which may be one of the mechanisms of triptolide immunosuppression and anti-inflammatory effects.
