目的:研究α-甘草次酸衍生物(GB)对大鼠外周血淋巴细胞体外活化的影响,探讨其是否对淋巴细胞活性存在抑制作用。方法:无菌分离大鼠外周血淋巴细胞并制成悬液,分别与不同浓度的GB预先孵育2 h,加入促分裂原伴刀豆蛋白A(ConA),继续孵育24 h后,ELISA法检测上清液中IL-4、IFN-γ含量。结果:2 mg·L~(-1)和1 mg·L~(-1)GB能显著降低ConA刺激下大鼠淋巴细胞产生的IL-4水平,而0.5 mg·L~(-1)GB则提高淋巴细胞产生的IL-4水平;2 mg·L~(-1)GB显著降低ConA刺激下大鼠淋巴细胞产生的IFN-γ水平,而1 mg·L~(-1)和0.5 mg·L~(-1)GB则提高淋巴细胞产生的IFN-γ水平。结论:给药剂量不同,GB对活化的大鼠外周血淋巴细胞作用不同,值得深入研究。
Objective: To investigate the effect of α-glycyrrhetinic acid derivatives (GB) on the activation of peripheral blood lymphocytes in rats and whether there is an inhibitory effect on the activity of lymphocytes. Methods: Peripheral blood lymphocytes were aseptically isolated and made into suspension. The cells were preincubated with different concentrations of GB for 2 h, and then added into mitogen ConA. The cells were incubated for another 24 h. Supernatant IL-4, IFN-γ content. Results: 2 mg · L -1 and 1 mg · L -1 GB could significantly reduce the level of IL-4 produced by ConA in rat lymphocytes, while 0.5 mg · L -1 GB Increased the level of IL-4 produced by lymphocytes; 2 mg · L -1 GB significantly decreased the level of IFN-γ produced by lymphocytes stimulated by ConA, while 1 mg · L -1 and 0.5 mg · L ~ (-1) GB increases the level of IFN-γ produced by lymphocytes. CONCLUSION: Different doses of GB can affect the activation of peripheral blood lymphocytes in rats, which deserves further study.
