目的:探讨非小细胞肺癌(non-small cell lung cancer,NSCLC)中垂体瘤转化基因(PTTG)和增殖细胞核抗原(PCNA)蛋白的表达相关性及其与肺癌发生发展之间的关系。方法:应用免疫组织化学SP法检测57例NSCLC组织及6例良性病变肺组织中PTTG和PCNA蛋白的表达。结果:NSCLC组织PTTG和PCNA蛋白的阳性表达率分别为87.72%和61.4%,在良性病变肺组织中均未检测到两者的表达。上述两种组织之间的PTTG和PCNA蛋白表达均有显著性差异(P<0.05)。PTTG蛋白表达与TNM分期和淋巴结转移密切相关(P<0.05),与患者的性别、年龄、肿瘤大小、组织学类型及分化程度无关(P>0.05)。PCNA蛋白表达与肿瘤分化程度有关(P<0.05),与患者的性别、年龄、肿瘤大小、组织学类型、TNM分期和淋巴结转移无关(P>0.05)。癌组织中PTTG和PCNA蛋白的表达呈显著正相关(r=0.306,P=0.005)。结论:PTTG和PCNA在非小细胞肺癌中高表达,两基因表达成显著正相关,二者在非小细胞肺癌的发生、发展中有协同作用,并可作为非小细胞肺癌生物学行为的指标。
Objective: To investigate the correlation between the expression of PTTG and PCNA in non-small cell lung cancer (NSCLC) and its relationship with the occurrence and development of lung cancer. Methods: Immunohistochemical SP method was used to detect the expression of PTTG and PCNA protein in 57 NSCLC tissues and 6 benign lung tissues. Results: The positive expression rates of PTTG and PCNA protein in NSCLC tissues were 87.72% and 61.4%, respectively. No expression was detected in the lung tissues of benign lesions. The expressions of PTTG and PCNA in the above two groups were significantly different (P <0.05). PTTG protein expression was closely related to TNM staging and lymph node metastasis (P <0.05), but not to sex, age, tumor size, histological type and degree of differentiation (P> 0.05). The expression of PCNA was correlated with the degree of tumor differentiation (P <0.05), but not with the gender, age, tumor size, histological type, TNM stage and lymph node metastasis (P> 0.05). There was a significant positive correlation between the expression of PTTG and PCNA in cancer tissues (r = 0.306, P = 0.005). Conclusion: PTTG and PCNA are highly expressed in non-small cell lung cancer (NSCLC). The expression of PTTG and PCNA is significantly and positively correlated with each other. Both of them have synergistic effects in the development and progression of non-small cell lung cancer and may be used as an indicator of biological behavior of NSCLC.
