目的研究大孔树脂纯化天山雪莲有效成分的工艺条件。方法以紫丁香苷、绿原酸和芦丁的吸附量和解吸率为评价指标,筛选树脂,并优化了吸附和洗脱条件。结果HPD400型树脂对天山雪莲各有效成分具有较好的吸附分离性能,适于纯化天山雪莲有效部位。最佳工艺条件为:雪莲样品溶液4BV,上样液的pH值为2~3,吸附流速1BV/h,吸附时间为12h,先用4BV水洗涤,再用30%、50%乙醇各6、4BV进行梯度洗脱,流速1BV/h,合并30%、50%乙醇洗脱液,即为目标产品。各有效成分的质量分数均提高了5倍左右。结论采用HPD400树脂分离纯化天山雪莲中有效成分是可行的。
Objective To study the process conditions of purifying the active ingredients of Sophora japonica from Macroporous resin. Methods The adsorption and desorption rates of syringic acid, chlorogenic acid and rutin were used as evaluation indexes to screen the resin and optimize the adsorption and elution conditions. Results HPD400 resin has good adsorption and separation properties for the active ingredients of Saussurea involucrata, which is suitable for the purification of effective parts of Saussurea tsaenia. The optimum conditions were as follows: the volume of sample was 4 BV, the pH value of sample solution was 2 ~ 3, the flow rate of adsorption was 1 BV / h, the adsorption time was 12 h, washed with 4BV water first, 4BV gradient elution, flow rate 1BV / h, combined 30%, 50% ethanol eluent, is the target product. The mass fraction of the active ingredients are increased about 5 times. Conclusion It is feasible to separate and purify the active ingredients from the Chinese yam by using HPD400 resin.
