目的:建立以高效液相色谱法测定对坐叶中木犀草素含量的方法。方法:色谱柱为Diamonsil C18(250mm×4.6mm,5μm),流动相为甲醇-0.04%磷酸溶液(60∶40),流速为1mL.min-1,柱温为35℃,检测波长为350nm。结果:木犀草素进样量在101.6~812.8ng范围内与峰面积积分值呈良好线性关系(r=0.9999);平均回收率为97.32%,RSD=0.9%(n=9)。结论:本方法简便、准确、分离效果好,可用于对坐叶的质量控制。 Objective: To establish a method for the determination of luteolin in sitting leaves by high performance liquid chromatography. Methods: The chromatographic column was Diamonsil C18(250mm×4.6mm, 5μm). The mobile phase was methanol-0.04% phosphoric acid solution (60∶40), the flow rate was 1mL.min-1, the column temperature was 35°C, and the detection wavelength was 350nm. RESULTS: There was a good linear relationship between the luteolin injection volume and the integrated value of the peak area in the range of 101.6-812.8 ng (r=0.9999); the average recovery was 97.32%, and the RSD was 0.9% (n=9). Conclusion: This method is simple, accurate, and has good separation effect. It can be used to control the quality of sitting leaves.
