目的探讨广西巴马长寿人群血清胰岛素样生长因子Ⅰ(IGF-Ⅰ)水平及其影响因素。方法 2008年5月采用随机抽样的方法在广西巴马长寿区抽取160名长寿老人(90~110岁)作为长寿组,另抽取一般地区健康青壮年人(20~50岁)102名作为青壮年对照组。测量其身高、体质量、腰围、血压、血糖、血脂等,应用酶联免疫吸附法检测血清IGF-Ⅰ水平,利用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法分析长寿老人胰岛素样生长因子Ⅰ受体(IGF-ⅠR)基因3174位点多态性,并对长寿老人血清IGF-Ⅰ水平的影响因素进行多元逐步回归分析。结果长寿组血清IGF-Ⅰ水平低于青壮年对照组〔(114.0±31.5)μg/L与(140.7±37.3)μg/L,F=6.321,P<0.05〕。长寿组中,GG、GA和AA基因型携带者血清IGF-Ⅰ水平分别为(115.3±30.4)μg/L、(112.1±34.5)μg/L及(113.2±27.3)μg/L,差异无统计学意义(F=1.030,P=0.386)。多元线性回归分析显示,血清IGF-Ⅰ与体质指数和高密度脂蛋白胆固醇(HDL-C)呈正相关,与性别、年龄、民族、臀围、腰围、血压、血糖、总胆固醇、三酰甘油和低密度脂蛋白胆固醇均无直线相关关系。结论广西巴马长寿人群血清IGF-Ⅰ水平明显低于青壮年人。长寿人群IGF-ⅠR基因3174位点多态性可能对其血清IGF-Ⅰ表达无影响,血清IGF-Ⅰ水平与体质指数及血清HDL-C水平呈正相关。
Objective To investigate the level of serum insulin-like growth factor Ⅰ (IGF-Ⅰ) and its influential factors in the population of longevity in Bama of Guangxi. Methods In May 2008, 160 longevity old people (90-110 years old) were selected as the longevity group in Changshou District, Bama, Guangxi by random sampling method. Another 102 healthy young adults (20-50 years old) in general areas were selected as young adults Control group. The body weight, body weight, waist circumference, blood pressure, blood glucose and blood lipids were measured. The level of serum IGF-Ⅰ was detected by enzyme linked immunosorbent assay (ELISA). The level of IGF-Ⅰ was detected by polymerase chain reaction- restriction fragment length polymorphism (PCR-RFLP) 3174 locus polymorphism of insulin-like growth factor receptor (IGF-ⅠR) gene in the elderly and the influencing factors of serum IGF-Ⅰ level in elderly people were analyzed by multivariate stepwise regression analysis. Results The level of serum IGF-Ⅰ in longevity group was lower than that in young and middle-aged control group (114.0 ± 31.5) μg / L and (140.7 ± 37.3) μg / L, F = 6.321, P <0.05). In the longevity group, the serum IGF-Ⅰ levels of GG, GA and AA genotype were (115.3 ± 30.4) μg / L, (112.1 ± 34.5) μg / L and (113.2 ± 27.3) μg / L respectively Significance (F = 1.030, P = 0.386). Multivariate linear regression analysis showed that serum IGF-I was positively correlated with body mass index and high density lipoprotein cholesterol (HDL-C), but not with gender, age, ethnicity, hip circumference, waist circumference, blood pressure, blood glucose, total cholesterol, triglyceride Low-density lipoprotein cholesterol no linear correlation. Conclusions The level of serum IGF-Ⅰ in longevity population of Bama in Guangxi is significantly lower than that in young adults. Long-lived population IGF-ⅠR gene 3174 polymorphism may have no effect on its serum IGF-Ⅰ expression, serum IGF-Ⅰ level and body mass index and serum HDL-C levels were positively correlated.
