目的观察银杏提取物EGb761对小鼠肺腺癌血管生成的影响。方法构建小鼠肺腺癌模型,用银杏提取物EGb761腹腔注射给药,在10d、20d、30d、40d观察小鼠的整体状态,计算肺组织肿瘤转移结节数目、瘤重及抑瘤率,检测肿瘤组织中血管内皮生长因子(VEGF)、肿瘤血管生长(MVD)、凝血酶敏感蛋白-1(TSP-1)的表达。结果 EGb761各剂量组的瘤重及肺转移灶数目均低于模型组,EGb761各剂量组VEGF表达、肿瘤血管密度均低于模型组,EGb761各组TSP-1显著增高,均具有显著性差异(P<0.05)。结论在小鼠肺腺癌模型中,银杏提取物EGb761可抑制小鼠肺腺癌肿瘤血管生成,其机制可能是通过调节VEGF表达减少,TSP-1表达增高而达到的。
Objective To observe the effect of ginkgo biloba extract EGb761 on angiogenesis in mouse lung adenocarcinoma. Methods The mouse lung adenocarcinoma model was established. The ginkgo biloba extract EGb761 was intraperitoneally injected to observe the overall status of mice at 10d, 20d, 30d and 40d. The number of tumor metastasis nodules, tumor weight and tumor inhibition rate were calculated. The expression of vascular endothelial growth factor (VEGF), tumor angiogenesis (MVD) and thrombospondin-1 (TSP-1) in tumor tissue were detected. Results The tumor weight and the number of lung metastases in EGb761 groups were lower than those in model group. The expression of VEGF and the density of tumor vessels in each group of EGb761 were lower than those in model group, and the levels of TSP-1 in EGb761 group were significantly increased P <0.05). Conclusion EGb761 can inhibit tumor angiogenesis in mouse lung adenocarcinoma in mice model of lung adenocarcinoma. The mechanism may be through reducing the expression of VEGF and increasing the expression of TSP-1.
