目的观察丁酸钠(SB)对急性白血病细胞共刺激分子表达的影响,并探讨其分子机制。方法以流式细胞术检测NB4、HL-60、Kasumi1、U937和Jurkat细胞经SB处理前、后CD86、CD80分子表达,同种混合淋巴细胞反应观察免疫调节功能,NFκB试剂盒检测SB对核内NFκB含量的影响。结果经SB处理后,各白血病细胞株细胞CD86和CD80分子表达与对照组比较均出现不同程度上调,其中0.5mmol/LSB处理的NB4细胞组较空白对照组CD86阳性细胞率增高36.8倍,NF-κB含量增加与CD86分子上调相一致,混合淋巴细胞反应显示SB处理的细胞可明显刺激异基因淋巴细胞增殖。结论SB可上调急性白血病细胞共刺激分子CD86和CD80,NF-κB为其参与上调CD86分子的重要的转录因子。
Objective To observe the effect of sodium butyrate (SB) on the expression of costimulatory molecules in acute leukemia cells and to explore its molecular mechanism. Methods The expressions of CD86 and CD80 on NB4, HL-60, Kasumi1, U937 and Jurkat cells before and after treatment with SB were detected by flow cytometry. The immunomodulatory function was observed by allogeneic mixed lymphocyte reaction. NFκB content. Results After treatment with SB, the expressions of CD86 and CD80 in all the leukemia cell lines were up-regulated compared with the control group. The percentage of CD86 positive cells in NB4 cells treated with 0.5mmol / LSB was 36.8 times higher than that in control cells, The increase of κB was consistent with the up-regulation of CD86 molecules. The mixed lymphocyte reaction showed that SB-treated cells could obviously stimulate allogeneic lymphocyte proliferation. Conclusion SB can up-regulate the costimulatory molecules CD86 and CD80 in acute leukemia cells. NF-κB is an important transcription factor involved in the up-regulation of CD86 molecules.
