目的:建立测定草乌中准噶尔乌头碱含量的方法。方法:采用反相高效液相色谱法,Agilent Extend-C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.1%三乙胺溶液(40∶60),流速1.0 m L·min~(-1),柱温25℃;蒸发光散射检测器(ELSD)的漂移管温度96℃,气体(空气)压力2.50 bar,撞击器呈关闭状态。结果:准噶尔乌头碱在1.5~30μg呈良好线性关系(r=0.999 5),平均回收率为96.86%,RSD 1.7%。结论:该方法简便、准确、快速,可用于测定草乌中准噶尔乌头碱的含量,为草乌的质量评价提供依据。
Objective: To establish a method for the determination of aconitine in Zuogar aconite. Methods: The mobile phase was acetonitrile-0.1% triethylamine (40:60) at a flow rate of 1.0 m L · min-1 using reversed-phase high performance liquid chromatography with Agilent Extend-C18 column (4.6 mm × 250 mm, (-1) and column temperature 25 ℃. The evaporating light scattering detector (ELSD) has a drift tube temperature of 96 ° C and a gas (air) pressure of 2.50 bar. The striker is turned off. Results: There was a good linear relationship between the concentration of aconitine and 1.5 ~ 30μg (r = 0.999 5). The average recoveries were 96.86% and RSD 1.7%. Conclusion: The method is simple, accurate and rapid, which can be used to determine the content of aconitine in Zhugu of Zhulgar, and provide the basis for the quality evaluation.
