目的:观察益肾Ⅰ方对多囊卵巢综合征(PCOS)大鼠胰岛素抵抗以及性激素的改善作用。方法:采用十一酸睾酮加绒促性素(HCG)建立PCOS大鼠模型,连续ig二甲双胍或不同剂量的益肾Ⅰ方6周,观察大鼠血清肿瘤坏死因子-α(TNF-α)、空腹血糖(FBG)、糖耐量(OGTT-2hBG)、胰岛素(Ins)含量和胰岛素敏感指数(ISI)、睾酮(T)、胰岛素样生长因子-Ι(IGF-1)、雌激素(E2)、孕酮(P)、泌乳素(PRL)的变化。结果:PCOS大鼠TNF-α,FBG,OGTT-2hBG和Ins水平以及T,IGF-1,E2和PRL含量明显高于对照组;ISI,P水平则显著降低(均P<0.01)。二甲双胍和益肾Ⅰ方3个剂量组均能不同程度地改善PCOS大鼠的上述病理改变(P<0.01~0.05),但益肾Ⅰ方低剂量组PRL和ISI未见明显改变(P>0.05)。结论:二甲双胍及益肾Ⅰ方均能不同程度地改善PCOS大鼠胰岛素抵抗以及性激素代谢异常。
Objective: To observe the effect of Yishen Ⅰ prescription on insulin resistance and sex hormones in rats with polycystic ovary syndrome (PCOS). Methods: PCOS rat model was established by using testosterone undecanoate plus cashmectin (HCG). Six consecutive weeks ig metformin or different doses of Yishen Ⅰ were used to observe the changes of serum tumor necrosis factor-α (TNF-α) FBG, OGTT-2hBG, Ins, ISI, T, IGF-1, Progesterone (P), prolactin (PRL) changes. Results: The levels of TNF-α, FBG, OGTT-2 hBG and Ins, the contents of T, IGF-1, E2 and PRL in PCOS rats were significantly higher than those in the control rats. The levels of ISI and P in PCOS rats were significantly decreased (all P <0.01). Both metformin and Yishen Ⅰ could significantly improve the above pathological changes in PCOS rats (P <0.01 ~ 0.05), but there was no significant change in PRL and ISI of Yishen Ⅰ low dose group (P> 0.05 ). Conclusion: Both metformin and Yishen Ⅰ can ameliorate insulin resistance and sex hormone metabolism in PCOS rats.
