目的探讨乌司他丁对大鼠肠缺血再灌注时肝组织Bcl-2、Bax蛋白表达的影响。方法成年健康Wistar大鼠24只,随机分为三组(n=8):假手术组(Sham组,S)、肠缺血再灌注组(intestine ischemical-reperfusion组,IIR)、乌司他丁组(ulinastatin组,UTI)。通过夹闭肠系膜上动脉1h后再灌注3h制作肠缺血再灌注损伤模型,采用免疫组化法与医学图像分析检测肝组织Bcl-2与Bax蛋白表达量。结果与S组比较,IIR组和UTI组Bcl-2与Bax蛋白含量均升高(P<0.05),IIR组Bcl-2/Bax比值降低(P<0.05),UTI组Bcl-2/Bax比值升高(P<0.05)。与IIR组比较,UTI组Bcl-2蛋白含量、Bcl-2/Bax比值均升高(P<0.05),Bax蛋白含量降低(P<0.05)。结论乌司他丁可能通过调节Bcl-2、Bax蛋白表达减轻肠缺血再灌注时肝的损伤。
Objective To investigate the effect of ulinastatin on the expression of Bcl-2 and Bax in liver tissue during intestinal ischemia-reperfusion in rats. Methods Twenty-four adult healthy Wistar rats were randomly divided into three groups (n = 8): sham operation group (Sham group, S), intestine ischemia-reperfusion group (IIR) Group (ulinastatin group, UTI). The intestinal ischemia-reperfusion injury model was made by clamping the superior mesenteric artery 1h and reperfusion 3h. The expression of Bcl-2 and Bax protein in liver tissues was detected by immunohistochemistry and medical image analysis. Results Compared with group S, the protein levels of Bcl-2 and Bax increased in IIR group and UTI group (P <0.05), but decreased in IIR group (P <0.05). The ratio of Bcl-2 / Bax Increased (P <0.05). Compared with IIR group, Bcl-2 protein content and Bcl-2 / Bax ratio increased (P <0.05) and Bax protein content decreased (P <0.05) in UTI group. Conclusion Ulinastatin may reduce the damage of liver during intestinal ischemia / reperfusion by regulating the expressions of Bcl-2 and Bax proteins.
