目的观察中成药坤宁安对围绝经期大鼠卵巢颗粒细胞凋亡的影响。方法将雌性13~15月龄大鼠50只随机分为模型组、坤宁安高、低剂量组、西药对照组、中药对照组,每组10只,另选5月龄雌性大鼠10只作青年对照组。建立围绝经期大鼠的模型,应用免疫组化检测方法观察不同研究组大鼠卵巢颗粒细胞中Bax蛋白的阳性表达,用原位末端标记法检测BaxmRNA的表达。结果Bax及BaxmRNA在模型组均呈强阳性表达,在青年对照组呈弱阳性表达,两组比较差异有统计学意义(P<0.01);而坤宁安高、低剂量及西药对照组,Bax及Baxm-RNA均呈弱阳性表达,与模型组比较,差异有统计学意义(P<0.01);中药对照组与模型组比较,差异无统计学意义(P>0.05)。结论①选用13~15月龄的wister雌性大鼠连续阴道涂片10d,选用持续动情大鼠作为围绝经期综合征的动物模型。实验证明此种造模方式可成功复制。②围绝经期大鼠卵巢颗粒细胞凋亡受凋亡促进基因Bax的影响,与Bax基因蛋白的阳性表达呈正相关。
Objective To observe the effect of KN on the apoptosis of ovarian granulosa cells in perimenopausal rats. Methods Fifty female rats aged 13-15 months were randomly divided into model group, Kunning Angao, low dose group, western medicine control group, traditional Chinese medicine control group, 10 rats in each group, and 10 female rats aged 5 months were selected. As a young control group. A model of perimenopausal rats was established. Immunohistochemistry was used to observe the positive expression of Bax protein in ovarian granulosa cells of different groups. The expression of Bax mRNA was detected by in situ end-labeling. Results The expression of Bax and Bax mRNA was strongly positive in the model group and weakly positive in the young control group. The difference between the two groups was statistically significant (P<0.01). The high and low dose of Baining and the western medicine control group were Bax. Both Baxm-RNA and weak Baxm-RNA expression were statistically significant compared with the model group (P<0.01). There was no significant difference between the Chinese medicine control group and the model group (P>0.05). Conclusion 1 Continuous vaginal smears of female wistar rats aged from 13 to 15 months were selected for 10 days. Continuous estrus rats were selected as an animal model of perimenopausal syndrome. Experiments have proved that this mode can be successfully copied. 2 The apoptosis of ovarian granulosa cells in perimenopausal rats was affected by the apoptosis-promoting gene Bax, which was positively correlated with the positive expression of Bax protein.
