将不同浓度梯度核糖核酸Ⅱ作用于7种细胞系,采用WST-8比色法测定其对细胞增殖的抑制情况,选择较敏感的人红白血病细胞K562作为试验对象,考察了细胞接种密度、药物作用时间等条件的影响,初步建立了注射用核糖核酸Ⅱ体外抗肿瘤活性的测定方法。核糖核酸Ⅱ在0.02~100 mg/ml范围内线性关系良好。采用本法测定了10批样品的抗肿瘤活性,结果对K562细胞的抑制率均大于55%。 Different concentrations of ribonucleic acid Ⅱ were applied to seven cell lines, and the inhibition of cell proliferation was measured by WST-8 colorimetric assay. K562, a sensitive human erythroleukemia cell line, was selected as the test object. The cell seeding density, Effect time and other conditions, initially established a method of determination of in vitro anti-tumor activity of ribonucleic acid for injection Ⅱ. Ribonucleic acid Ⅱ showed good linearity in the range of 0.02-100 mg / ml. The antitumor activity of 10 batches of samples was determined by this method. The results showed that the inhibition rate of K562 cells was more than 55%.
