目的从原代培养获得角膜缘干细胞(LSCs),探讨不同浓度胰岛素对角膜缘干细胞增殖作用的影响。方法取兔角膜组织体外培养获得角膜缘干细胞,并行免疫组织化学及透射电镜鉴定。5mg/L、10mg/L、20mg/L、40mg/L、80mg/L及100mg/L胰岛素作用后,以四甲基偶氮唑盐法检测胰岛素对角膜缘干细胞增殖能力的影响。结果 5mg/L和10mg/L浓度的胰岛素有促进角膜缘干细胞增殖的作用,与对照组比较有显著性差异(P<0.05);5mg/L与10mg/L组比较也有显著差异(P<0.05);20~100mg/L组与对照组比较无显著性差异(P>0.05)。结论胰岛素促进培养角膜缘干细胞的增殖,最适宜浓度为5mg/L。
Objective To investigate the effects of different concentrations of insulin on the proliferation of limbal stem cells (LSCs) derived from primary culture. Methods The corneal limbal stem cells were obtained by culturing rabbit corneas in vitro. Immunohistochemistry and transmission electron microscopy were performed. The effects of insulin on the proliferation of limbal stem cells were detected by MTT assay after 5 mg / L, 10 mg / L, 20 mg / L, 40 mg / L, 80 mg / L and 100 mg / L insulin respectively. Results Insulin at concentrations of 5 mg / L and 10 mg / L enhanced the proliferation of limbal stem cells, which was significantly different from that of the control group (P <0.05). There were also significant differences between the 5 mg / L and 10 mg / L groups ); 20 ~ 100mg / L group compared with the control group no significant difference (P> 0.05). Conclusion Insulin promotes the proliferation of cultured limbal stem cells, the most suitable concentration is 5mg / L.
