目的研究TNF-α基因-308G/A多态性与中国人群冠状动脉粥样硬化性心脏病(coronary atherosclerotic heart disease,CHD)的关系。方法计算机检索Pub Med、Web of Science、CNKI、Wan Fang Data和VIP数据库,搜集TNF-α基因-308G/A多态性与中国人群CHD相关的病例-对照研究,检索时限均为建库至2017年2月。由两名研究者独立地进行文献筛选、资料提取和评价纳入研究的偏倚风险评价后,采用Stata 12.0软件进行Meta分析。结果最终纳入10个病例-对照研究,包括4 628例患者和4 362例对照人群。Meta分析结果显示:TNF-α基因-308G/A多态性与中国人群CHD易感性之间存在相关性[A vs.G:OR=1.13,95%CI(1.02,1.26),P=0.020;AA vs.GA/GG:OR=1.47,95%CI(1.02,2.12),P=0.038;AA vs.GG:OR=1.50,95%CI(1.04,2.16),P=0.029]。结论 TNF-α基因-308G/A多态性与中国人群CHD易感性之间存在相关性,A等位基因为其潜在危险因素。受纳入研究的数量与质量限制,上述结论尚需开展更多高质量研究予以验证。
Objective To investigate the relationship between TNF-α gene-308G / A polymorphism and coronary atherosclerotic heart disease (CHD) in Chinese population. Methods PubMed, Web of Science, CNKI, Wan Fang Data and VIP databases were searched by computer. The case-control study of TNF-α gene-308G / A polymorphism in Chinese population was collected. In February. Two researchers independently conducted literature screening, data extraction and evaluation were included in the study after the risk assessment of bias, using Stata 12.0 software for meta-analysis. The results eventually included 10 case-control studies, including 4,628 patients and 4,362 controls. Meta-analysis showed that there was a correlation between -308G / A polymorphism of TNF-α gene and CHD susceptibility in Chinese population [A vs. G: OR = 1.13, 95% CI 1.02,1.26, P = 0.020; AA vs. GA / GG: OR = 1.47,95% CI (1.02,2.12), P = 0.038; AA vs.GG: OR = 1.50,95% CI (1.04,2.16), P = 0.029]. Conclusion There is a correlation between -308G / A polymorphism of TNF-α gene and CHD susceptibility in Chinese population. A allele is a potential risk factor. Due to the quantity and quality limitations of the included studies, the above conclusion still needs to be verified by more high-quality research.
