固体分散体可用于增加或减小药物的溶出速率。能加速药物溶出的分散体,其制法及作用机制已有详尽综述。减小溶出速率的方法通常是将药物混合于不溶性载体如乙基纤维素及蜡中来达到。这种类型的固体分散体有时可看作是骨架体系(Matrix system)。该体系能延长药物的释放时间,因此适用于设计缓释剂型,它具有减少用药次数及改善病人顺应性等优点。
Solid dispersions can be used to increase or decrease the dissolution rate of a drug. Dispersions that accelerate the dissolution of drugs have been reviewed in detail for their formulation and mechanism of action. The method of decreasing the dissolution rate is usually achieved by mixing the drug in an insoluble carrier such as ethyl cellulose and wax. This type of solid dispersion can sometimes be seen as a matrix system. The system can prolong drug release time and is therefore suitable for the design of sustained release dosage forms which have the advantage of reducing the number of dosing and improving patient compliance.
