目的探讨枸杞中甜菜碱、牛磺酸、枸杞多糖(lycium barbarum polysaccharides,LBP)分别对β-胡萝卜素(β-carotene,β-C)在比格犬体内药动学的影响。方法采用随机、对照双周期交叉实验设计,将6只比格犬随机分为2组,分别给予β-胡萝卜素,β-胡萝卜素合并甜菜碱或牛磺酸或枸杞多糖;按设定时间点采血,采用HPLC测定比格犬血浆中β-胡萝卜素、视黄醇、视黄醉棕榈酸酯的血药浓度,经DAS2.0.1计算药动学参数。结果与单独给β-胡萝卜素相比,合并给药后甜菜碱可使β-胡萝卜素在比格犬体内t_(1/2)显著延长(P<0.05);牛磺酸可增大β-胡萝卜素的AUC_(0-t),延长其MRT,同时降低清除率(P<0.05);而LBP对β-胡萝卜素在比格犬体内的经时过程无显著性影响(P>0.05)。结论枸杞中甜菜碱和牛磺酸可显著影响β-胡萝卜素在比格犬体内的药动学,表明枸杞“滋肝明目”并不是一种成分单一作用的结果,而是多种成分共同作用的结果。
Objective To investigate the effects of β-carotene (β-C) on the pharmacokinetics of beagle dogs in the beagle dogs of betaine, taurine and lycium barbarum polysaccharides (LBP). Methods A randomized, controlled two-cycle crossover design was used. Six Beagle dogs were randomly divided into two groups, and were given β-carotene, β-carotene combined with betaine or taurine or LBP respectively. Blood samples were collected for determination of β-carotene, retinol and retinyl palmitate in beagle dogs by HPLC. The pharmacokinetic parameters were calculated by DAS 2.0.1. Results Compared with β-carotene alone, β-carotene was significantly prolonged (t 1/2) in beagle dogs (P <0.05), while taurine increased β- The carotene AUC_ (0-t) prolonged its MRT and decreased the clearance rate (P <0.05). However, LBP had no significant effect on the time course of β-carotene in beagle dogs (P> 0.05). Conclusion The betaine and taurine in wolfberry can significantly affect the pharmacokinetics of β-carotene in Beagle dogs, indicating that wolfberry “Zi-Gan Zhi-Mu” is not the result of a single role of the composition, but a variety of ingredients Result of joint action.
