小麦高分子量麦谷蛋白亚基(HMW-GS)组成与其加工品质密切相关,建立其相关简便有效的遴选体系对小麦品质评价和优质品种选育具有重要意义。本研究选用对小麦品质有重要影响的HMW-GS基因的特异性分子标记,基于26份已知HMW-GS组成的小麦品种(系)检测验证基础上,建立强筋小麦HMW-GS基因的多重PCR检测体系。已建立了4套多重PCR体系:体系Ⅰ同时检测Axnull与By8基因,体系Ⅱ检测By8与Dx5基因,体系Ⅲ同时检测Axnull、By8与Dx5基因,体系Ⅳ同时检测Axnull、Bx14与Dx5基因。利用每个体系对26份品种(系)进行检测,其结果与已知结果一致,说明创建的4套多重PCR体系稳定结果可靠,可用于强筋小麦品种的分子辅助选择和品质评价。
The composition of wheat high molecular weight glutenin subunit (HMW-GS) is closely related to its processing quality. It is of great importance to establish a simple and effective selection system for wheat quality evaluation and breeding of high quality varieties. Based on the testing and verification of 26 wheat cultivars (lines) composed of 26 known HMW-GSs, the HMW-GS gene of strong gluten wheat was identified as multiple molecular markers PCR detection system. Four sets of multiplex PCR systems have been established: system Ⅰ detects both Axnull and By8 genes simultaneously, system Ⅱ detects By8 and Dx5 genes, system Ⅲ simultaneously detects Axnull, By8 and Dx5 genes, and system Ⅳ simultaneously detects Axnull, Bx14 and Dx5 genes. Twenty-six varieties (lines) were tested with each system. The results were consistent with the known results. The results showed that the four multiplex PCR systems were stable and reliable, which could be used for molecular aided selection and quality evaluation of strong gluten wheat varieties.
