生理药物代谢动力学模型(PBPK模型)是当前药效研究和毒理学研究的重要方法。不同于传统的实验方法及经验模型,PBPK模型是一种基于机制的数学模型,用于模拟化学物质在体内的分布代谢,并且方便进行各种外推。该文主要总结了当前PBPK模型及其建立的一般步骤和方法,介绍了复合生理药物代谢动力学模型,并对生理药物代谢动力学模型在毒理学领域和健康风险评价中的应用进行了探讨,指出了PBPK模型发展受到的限制和未来的发展前景。
Physiological pharmacokinetic model (PBPK model) is an important method for the current pharmacodynamic and toxicological studies. Unlike the traditional experimental methods and empirical models, the PBPK model is a mechanism-based mathematical model used to model the distributional metabolism of chemicals in the body and facilitates various extrapolations. This paper summarizes the current PBPK model and its general steps and methods, introduces the compound physiologic pharmacokinetic model, and discusses the application of the physiological pharmacokinetic model in the field of toxicology and health risk assessment. The limitation of the development of PBPK model and its future development are pointed out.
