目的探讨慢性髓细胞性白血病(CML)的基因表达变化。方法分别从正常人和CML患者骨髓样品中抽取骨髓单个核细胞,分离出cDNA片段,然后再逆转录成cRNA并标记后,与人的全基因组表达谱芯片杂交,ABI1700芯片分析系统进行分析基因表达谱的差异。结果总共发现差异表达的基因有6706个,其中与CML相关的差异基因有75个(上调19个,下调56个)。结论全基因组寡核苷酸基因芯片在分析基因表达谱差异研究中具有广泛的应用价值。
Objective To investigate the gene expression of chronic myelogenous leukemia (CML). Methods Bone marrow mononuclear cells were extracted from bone marrow samples of normal and CML patients, respectively. The cDNA fragments were isolated and then reverse transcribed into cRNA and labeled. Then, human bone marrow mononuclear cells were hybridized with human genome-wide expression microarray. ABI1700 chip analysis system was used to analyze gene expression Differences in spectra. Results A total of 6706 differentially expressed genes were found, of which 75 were related to CML (19 up-regulated and 56 down-regulated). Conclusion The genome-wide oligonucleotide microarray has a wide range of applications in the analysis of gene expression profiles.
