目的:探讨不同剂量玉米油与猪油的1∶1调和油对小鼠PPARα、PPARγ、RXRα基因表达的影响。方法:将24只KM鼠随机分为3组,分别饲喂不同油脂含量的饲料,即3.8%推荐摄入量组(B组)、6.5%实际摄入量组(C)及对照组(A)。持续饲养8周并于每周一上午9:00称量每只小鼠体质量,8周后,采集肝脏组织,测定肝脏中的PPARα、PPARγ、RXRαm RNA表达水平。结果:小鼠体增长率与调和油剂量呈正相关;推荐摄入量组和实际摄入量组PPARαm RNA表达量均极显著低于对照组;推荐摄入量组PPARγm RNA表达量极显著高于对照组,实际摄入量组PPARγm RNA表达量极显著低于推荐摄入量组,实验组RXRαm RNA表达量与对照组比较无显著性差异。结论:每天摄入25 g玉米油和猪油的1∶1调和油能促进PPARγm RNA的表达,有效抑制非酒精性脂肪肝的形成。
Objective: To investigate the effect of different doses of corn oil and lard oil 1: 1 blend oil on PPARα, PPARγ and RXRα gene expression in mice. Methods: Twenty-four KM mice were randomly divided into three groups and fed with different fat content (group B, 3.8%), control group (6.5% ). After 8 weeks of continuous feeding, the body weight of each mouse was weighed at 9:00 am every Monday. After 8 weeks, liver tissues were harvested and the expression of PPARα, PPARγ and RXRαmRNA in the liver was measured. Results: There was a positive correlation between the growth rate of mice and the dosage of reconstituted oil. The PPARαm RNA expression levels in the recommended intake and actual intake groups were significantly lower than those in the control group. The PPARγm RNA expression level in the recommended intake group was significantly higher than that in the control group In the control group and the actual intake group, the expression of PPARγm RNA was significantly lower than the recommended intake group. There was no significant difference in the expression level of RXRαmRNA in the experimental group compared with the control group. Conclusion: Daily intake of 25 g corn oil and lard oil 1: 1 blend oil can promote the expression of PPARγm RNA, effectively inhibit the formation of non-alcoholic fatty liver.
