目的:探讨血细胞分离机分离的外用血造血干细胞经程序降温冷冻保存、复温后NC、MNC、台盼兰拒染的回收效果。方法:应用CS-3000PLUS血细胞分离机对52例成人、儿童急性、非急性淋巴细胞性白血病、非何杰金氏淋巴瘤等患者进行了100例次分离,采集的外周血造血干细胞悬液用二步降温法进行程序降温冷冻保存。结果:在液氮中保存6~78天的外周血经40~45℃水浴中快速复温NC、MNC、台盼兰拒染回收率分别为107.3%、94.4%、95.1%。结论:用血细胞分离机分离的外周血干细胞再进行冻存,减少细胞损伤,得到数量较多和质量保证的干细胞;可以重复进行,缩小体积,方便冷冻处理及保存;减少污染机会,提高工作效率。
OBJECTIVE: To investigate the recovery effect of blood-free hematopoietic stem cells isolated from blood cell separator after cryopreservation and rewarming of NC, MNC, and trypan blue. METHODS: One hundred and twenty patients with adult, childhood acute, non-acute lymphocytic leukemia and non-Hodgkin’s lymphoma were divided into two groups by CS-3000PLUS blood cell separator. Peripheral blood stem cell suspension was collected with two methods. Step cooling method for program cooling and cryopreservation. RESULTS: The rapid recovery of peripheral blood from 6 to 78 days in liquid nitrogen after incubation in a 40-45°C water bath with NC, MNC, and trypan blue was 107.3%, 94.4%, and 95.1 respectively. %. Conclusion: Peripheral blood stem cells isolated by blood cell separators are cryopreserved to reduce cell damage, and stem cells are obtained in larger quantity and quality assurance; they can be repeated, reduced in size, convenient for freezing and preservation, and reduce the chance of contamination and improve work efficiency. .
