目的 探讨粉防己碱诱导人红白血病细胞 (K56 2 )凋亡的作用。 方法 采用光镜、电镜、免疫荧光观察K56 2 细胞形态的改变 ,以流式细胞仪分析细胞周期 ,以ABC法检测细胞中Bcl 2和p5 3蛋白的改变 ,以TUNEL法检测细胞凋亡。 结果 K56 2 细胞经粉防己碱诱导 4 8h后 ,出现细胞凋亡早期形态学改变 ;流式细胞仪显示细胞DNA合成受到抑制 ;ABC法检测出细胞中Bcl 2水平降低和p5 3水平升高 ;TUNEL法原位检测揭示有DNA断裂。结论 粉防己碱可抑制K56 2 细胞的生长 ,其作用与浓度相关 ,最终可导致细胞凋亡
Objective To investigate the effect of tetrandrine on apoptosis of human erythroleukemia cells (K56 2). Methods The morphological changes of K56 2 cells were observed by light microscope, electron microscope and immunofluorescence. The cell cycle was analyzed by flow cytometry. The changes of Bcl 2 and p 5 3 proteins were detected by ABC method. The apoptosis was detected by TUNEL method. Results The morphological changes of K56 2 cells induced by tetrandrine at early stage were observed. Flow cytometry showed that the cell DNA synthesis was inhibited. The levels of Bcl 2 and p5 3 in the cells were decreased by ABC method. In situ detection by TUNEL revealed DNA fragmentation. CONCLUSION Tetrandrine inhibits the growth of K56 2 cells and its effect is related to its concentration and eventually leads to apoptosis
